Preclinical experiments have shown that AS1402 can act as a 'marker flag', attaching itself to tumour cells and helping components of the patient’s immune system to find and destroy them.
huHMFG1 binds specifically to MUC1, a cell membrane protein present in a variety of tumours, including breast, ovarian, pancreatic, gastric and colon cancers. Expression of MUC1 is generally associated with poor prognosis. In laboratory experiments, huHMFG1 has been shown to be a potent inducer of antibody-dependent cellular cytotoxicity (ADCC).
Breast cancer
AS1402 has completed a phase I dose escalation study in 26 patients with locally advanced or metastatic breast cancer who had relapsed following chemotherapy. AS1402 was well tolerated and no maximum tolerated dose (MTD) was reached. Some cases of stable disease were observed. The drug will now progress into a phase II study in advanced, metastatic breast cancer.
